Charles River Becomes Exclusive Global Provider of p53 and Bcrp TGEM™ Knockout Rat Models

Charles River has entered into an exclusive agreement with Transposagen Biopharmaceuticals Inc., a leading rat knockout model creation company, to be the sole distributor of their p53 and Bcrp TGEM™ rat models.  Transposagen has teamed with the colony management expertise of Charles River to offer these models to the research community.

Tumor protein 53 (Tp53, p53) is the most studied tumor suppressor. The p53 plays a role in cell cycle control, apoptosis, angiogenesis, carcinogenesis, senescence, DNA repair and changes in metabolism. Homozygous and heterozygous p53 TGEM™ rats develop a wide variety of malignant tumors, including sarcomas and lymphomas.

The p53 activates many genes that are thought to produce a comprehensive antioxidant effect, and p53 expression controls the level of aging-associated oxidative damage. The p53 TGEM™ rat model may be useful for aging and antioxidant effects.

The p53 pathway is a suppressor of tumorigenicity and also induced pluripotent stem (iPS) cell generation. Many rodent embryonic fibroblasts lacking p53 expression generate iPS cells more efficiently than wild-type fibroblasts, and siRNA p53 knockdown promotes human iPS cell formation. Many p53-regulated genes in embryonic fibroblasts are common to both humans and rodents

As a member of the ABC-transporter family, Bcrp regulates the bioavailability of many pharmaceuticals and the Bcrp TGEM™ rat model will provide a convenient means of conducting in vivo transporter and drug-drug interaction studies.

Bcrp has a wide variety of functions. In addition to its role in drug transport, Bcrp also regulates the fate and function of various subpopulations of stem or progenitor cells, and the Bcrp TGEM™ rat model may be useful for in vivo assessment of stem cell therapies. Other roles include regulation of hypoxia through binding and transport of heme and involvement in phototoxicity caused by interruption of metabolic pathways of dietary products.
