Overview Technical Resources OriginTransgenic mice were produced by injecting an 11.8 kb (NotI fragment) human genomic GLUT 4 transgene including a 5.3 kb of 5’flanking sequence and an SV40 polyadenylation sequence into the pronuclei of embryos from matings of C57BL/KsJ-m+/+ males and superovulated females. The embryos were then transferred to pseudopregnant CD-1 females. Founder mice were identified by PCR of total genomic DNA to obtain two mice with the recessive obese phenotype (+db/+db) and two founders with the recessive misty coat color (m+/m+). Independent lines were established by backcrossing lean founder mice with C57BL/KsJ-m+/db+ mates. Transferred to Charles River under exclusive license in 2011.
