Stroke & Brain Ischemia Models

Charles River conducts proof-of-principle/proof-of-concept studies for neuroprotection and reduction of infarct, as well as longer-term, more comprehensive studies looking at functional and cognitive outcomes or stroke prevention. All studies can be customized and adjusted to meet client needs.

Animal models of stroke and brain ischemia available for use in our studies include:

  • Transient middle cerebral artery occlusion (tMCAO)
  • Permanent middle cerebral artery occlusion (MCAO)
  • Endothelin-1-induced middle cerebral artery occlusion (EMCAO)
  • Global brain ischemia
  • Hind limb ischemia peripheral model
  • Stroke prevention model in Dahl/Salt Sensitive (Dahl/SS) or Stroke-Prone Spontaneously Hypertensive (SHRSP) rats
  • Photothrombotic stroke model
Gd-enhanced MRI

Gadolinium (Gd)-enhanced MRI of an ischemic brain shows the magnitude of blood brain barrier (BBB) leakage and Gd accumulation to the infarcted hemisphere. Time series images (from left upper corner to right lower corner) acquired by T1-MRI after IV administration of Gd contrast agent.

T2 and Diffusion Imaging

Absolute T2 and diffusion imaging at 24 and 48h after stroke in rats. These MRI techniques allow evaluation of brain tissue viability (absolute T2 values) and edema (diffusion) in an ischemic brain. These imaging modalities extend the value of MRI beyond typical assessment of ischemic injury (infarct volume) by enhancing understanding of stroke pathology and effects of novel therapies in cerebrovascular disease models.
