Charles River Introduces THE POUND MOUSE™

In 2005, a technician at our Kingston, NY facility noticed animals in a C57BL/6NCrl colony that appeared to be obese. The affected animals, as well as their parents, siblings and relatives, were identified, removed from the colony, and transferred to our Wilmington facility for characterization.

Based on the initial phenotype, circulating leptin levels, and complementation studies, our DNA analysis focused on the leptin receptor gene. DNA sequence analysis revealed an exon 2 deletion in the leptin receptor gene, likely resulting in complete lack of the leptin receptor in mice homozygous for this mutation. The obesity mutation found in this colony is in the same gene as that found in the BKS.Cg-m+/+Leprdb/J (db) mouse.

Nomenclature for this mutant mouse strain has been established and registered as C57BL/6N-Leprdb-lb/Crl, though the model is more commonly referred to as THE POUND MOUSE™. Male and female mutants are sterile, so a PCR assay supports selection of heterozygote breeders. THE POUND MOUSE™ is a true coisogenic strain; a spontaneous mutation has occurred on the popular C57BL/6NCrl background. C57BL/6NCrl mice do not carry the Nnt gene mutation, a mutation associated with dysfunctional glucose metabolism.

This model will be valuable to researchers studying the risk factors associated with metabolic syndrome, specifically obesity, insulin resistance, dyslipidemia, and glucose intolerance. The genetic defect in the leptin receptor of THE POUND MOUSE™ is also expected to facilitate pathophysiology studies of metabolic syndrome. In addition, we expect the C57BL/6NCrl-Leprdb-lb/Crl mouse model to be of value to scientists studying diseases commonly associated with metabolic syndrome, such as diabetes, cardiovascular disease, renal function, and non-alcohol-related fatty liver disease.
