Sample Preparation Procedure for Bacterial & Fungal Identification

Sample procedures are outlined in Sample Preparation Procedure (MGXOP-003).

Samples may be sent to Microgenetix for sequencing identification using one of two methods:

  • 1 (a). Agar Plates
    • Samples should be preferably grown on solid media such as agar plates.
    • Plates should be streaked for purity and single colonies.
    • Plate samples should be sealed tightly with laboratory film.
    • Use sturdy package materials to withstand breakage and/or leakage under normal shipping conditions.
    • All samples must be clearly labelled and submitted along with a completed Sequencing Identification Request Form or MALDI-TOF Identification Request Form.
    • All sections relating to customer and test information should be completed to expediate the testing of the samples.
  • 1 (b). Liquid Samples
    • Broth/liquid culture samples can be submitted to Microgenetix directly.
    • All liquid samples should be sealed tightly with laboratory film.
    • Use sturdy package materials to withstand breakage and/or leakage under normal shipping conditions.
    • All samples must be clearly labelled and submitted along with a completed Sequencing Identification Request Form or MALDI-TOF Identification Request Form.
    • All sections relating to customer and test information should be completed to expediate the testing of the samples.
    • It must be specified by the customer as to whether the liquid culture is to be transferred onto solid culture media by Microgenetix and purity checked or the sample is to be prepared directly for gene sequencing. It must be noted that sequencing samples where the purity of the sample is not known may result in an identification failure.
  • 2. Prepman Ultra Buffer

    Samples can be provided to Microgenetix in Prepman Ultra Buffer (Cat no. MGXPMAN-005/010/050/100). The preparations of these samples are detailed as follows:

    Plates

    • Select a single colony from the agar plate sample.
    • Using a fine sterile loop or wire touch a single colony of bacteria or filamentous fungi.
    • Transfer the inoculum to 200ul of Prepman Buffer (MGXPMAN).
    • All samples must be clearly labelled and submitted along with a completed Sequencing Identification Request Form or MALDI-TOF Identification Request Form.
    • All sections relating to customer and test information should be completed to expediate the testing of the samples.

    Liquid Samples

    • Pipette 1 ml of liquid culture samples into a sterile microcentrifuge tube.
    • Spin the sample for 3 minutes @ 16,000 rpm.
    • Aspirate and discard the supernatant.
    • Transfer a small amount of the pellet to 200ul of Prepman Buffer (MGXPMAN).
    • All samples must be clearly labelled and submitted along with a completed Sequencing Identification Request Form or MALDI-TOF Identification Request Form.
    • All sections relating to customer and test information should be completed to expediate the testing of the samples.

As part of the genetic sequencing service provided by Microgenetix, it is important to provide pure culture samples. Samples provided with mixed cultures may not allow for definitive identification to be provided. 

Samples can be sent to Microgenetix by post or courier. Please notify Microgenetix upon sending the samples so that they can be logged and included into the next available sequencing run.
